Neuro-oncology Multidisciplinary Clinic and Improvements in Patient Outcome
Background: Neuro-oncology care is becoming increasingly complex and patients often see multiple specialists. Multidisciplinary clinic (MDC) is a patient-centric option to allow multiple specialists to be involved where patients see multiple providers on the same day. Objective: The objective of this study was to evaluate the role of a neuro-oncology MDC on patient outcomes. Materials and Methods: Retrospective study was performed on consecutive patients who received radiosurgery...

Radiotherapy in Early-stage Gastric MALT: Improved Survival Without Increased Cardiac Death
Purpose: Radiotherapy (RT) is an effective treatment for localized gastric mucosa-associated lymphoid tissue (MALT) lymphomas unresponsive to antibiotic therapy; however, irradiating the stomach can result in significant radiation to the heart, a risk factor for cardiac disease. We analyzed the Surveillance, Epidemiology, and End Results database to evaluate outcomes related to cardiac disease among patients treated with RT for stage I gastric MALT. Materials and Methods: We identified...

PIK3CA mutation in the ShortHER randomized adjuvant trial for patients with early HER2+ breast cancer: association with prognosis and integration with PAM50 subtype.
Purpose: We explored the prognostic effect of PIK3CA mutation in HER2-positive patients enrolled in the ShortHER trial. Experimental Design: The ShortHER trial randomized 1253 patients with HER2-positive breast cancer to 9-weeks or 1-year of adjuvant trastuzumab combined with chemotherapy. PIK3CA hot-spot mutations in exon 9 and 20 were analysed by Pyrosequencing. Expression of 60 genes, including PAM50 genes was measured using the nCounter platform. Results: A mutation of the PIK3CA gene was detected...

Radiotherapy After Breast Conserving Surgery for Elderly Patients with Early‐Stage Breast Cancer: A National Registry Based Study
Abstract Considerable controversies exist regarding whether elderly patients with early‐stage breast cancer receiving breast conserving surgery (BCS) should forgo radiotherapy. We utilized the National Cancer Database to analyze data of 115 516 women aged ≥70 years, treated with BCS for T1‐2N0‐1M0 breast cancer between 2004 and 2014. Multivariable Cox proportional hazards model was used to estimate hazard ratios (HR) and 95% confidence intervals (CI) for mortality 3‐, 5‐ and 10‐years after 90‐days...

Toenail selenium, plasma selenoprotein P and risk of advanced prostate cancer: a nested case‐control study
Abstract Low selenium status may be associated with increased risk of prostate cancer (PC), particularly aggressive PC, and variation in selenoprotein genes may constitute an important modifying factor. We aimed to investigate the association between two selenium status biomarkers [toenail selenium, plasma selenoprotein P (SELENOP)] and risk of advanced, high‐grade, and advanced‐stage PC. We further studied whether variations in selenoprotein genes were associated with PC risk and selenium biomarker...

Metastatic melanoma patient outcomes since introduction of immune checkpoint inhibitors in England between 2014 and 2018
Abstract Immune checkpoint inhibitors (CPIs) have radically changed outcomes for patients diagnosed with metastatic melanoma globally in the last 10 years, based on evidence of overall survival (OS) benefits generated from international randomised controlled trials (RCTs). Since RCTs don’t always reflect real‐world prescribing, we interrogated established national databases to track prescribing of CPIs approved for first line treatment of metastatic melanoma patients in England since 2014 and determined...

Primary Care Providers’ Knowledge, Attitudes, Beliefs, and Practice Related to Lung Cancer Screening in Five High-Risk Communities in New York City
Abstract Racial/ethnic minorities face stark inequalities in lung cancer incidence, treatment, survival, and mortality compared with US born non-Hispanic Whites. Lung cancer screening (LCS) with low-dose computed tomography (LDCT) is effective at reducing lung cancer mortality in high-risk current and former smokers and is recommended by the US Preventive Services Task Force (USPSTF). This study sought to assess primary care providers’ (PCPs’) knowledge, attitudes, beliefs, and...

A Not So Perfect Score: Factors Associated with the Rate of Straight Line Scoring in Oncology Training Programs
Abstract Straight line scoring (SLS), defined as trainee assessments with the same score for all evaluation items, is statistically improbable and potentially indicates inaccurate assessment. Factors contributing to higher SLS rates are unknown, and knowledge of SLS prevalence within oncologic training is lacking. SLS frequency was measured for evaluations from all Accreditation Council for Graduate Medical Education (ACGME)-accredited programs at a single cancer care institution...

SALL4 promotes tumor progression in breast cancer by targeting EMT
Abstract Sal‐like protein 4 (SALL4) is overexpressed in breast cancer and might contribute to breast cancer progression, but the molecular mechanism remains unknown. Here, we found that within a group of 371 ethnic Chinese breast cancer patients, SALL4 was associated with lower grade (P = .002) and progesterone receptor positivity (P = .004) for overall cases; lower Ki67 (P = .045) and high vimentin (P = .007) for luminal cases. Patients with high SALL4 expression in lymph node metastasis showed...

A combined biological and clinical rationale for evaluating metastasis directed therapy in the management of oligometastatic prostate cancer
The past decade has seen the standard of care (SoC) for metastatic prostate cancer (MPC) expand beyond lifelong androgen deprivation therapy (ADT). Multiple randomized control trials (RCTs) have demonstrated improvements in overall survival by intensifying systemic therapy [1-8]. Such care has produced excellent outcomes for an advanced malignancy. In the most recent RCTs, newly diagnosed metastatic hormone sensitive prostate cancer (mHSPC) patients have 3-year overall survival rates between 65-80%.

Radiation-induced risk of ischemic heart disease following breast cancer radiotherapy in Denmark, 1977-2005
Over the past 15 years, evidence has accumulated that radiotherapy for breast cancer can increase the risk of subsequent heart disease. Parts of this evidence come from studies comparing left- with right-sided radiotherapy. Since the heart is located in the left side of the chest, the radiation dose to the heart tends to be larger for left- than for right-sided breast cancer. An increased risk of heart disease comparing left- with right-sided radiotherapy has been interpreted as evidence of radiation-induced...

In vitro studies provide insight into effects of Dicer-2 helicase mutations in Drosophila melanogaster [Article]
In vitro, Drosophila melanogaster Dicer-2 (Dcr-2) uses its helicase domain to initiate processing of dsRNA with blunt (BLT) termini, and its Platform•PAZ domain to initiate processing of dsRNA with 3’ overhangs (ovrs). To understand the relationship of these in vitro observations to roles of Dcr-2 in vivo, we compared in vitro effects of two helicase mutations to their impact on production of endogenous and viral siRNAs in flies. Consistent with the importance of the helicase domain in processing...

A rare bacterial RNA motif is implicated in the regulation of the purF gene whose encoded enzyme synthesizes phosphoribosylamine [Article]
The Fibro-purF motif is a putative structured noncoding RNA domain that was discovered previously in species of Fibrobacter by employing comparative sequence analysis methods. An updated bioinformatics search yielded a total of only 30 unique-sequence representatives, exclusively found upstream of the purF gene that codes for the enzyme amidophosphoribosyltransferase. This enzyme synthesizes the compound 5-phospho-D-ribosylamine (PRA), which is the first committed step in purine biosynthesis. The...
