On the use of actigraphy in clinical evaluation of diurnal blood pressure profile
Abstract A disturbed diurnal blood pressure profile is one of the most important risk factors of cardiovascular diseases. This review analyzes the use of simultaneous diurnal ambulatory blood pressure monitoring (ABPM) and motion activity monitoring (actigraphy) to obtain additional information for correct interpretation of ABPM results in clinically significant decision-making. The article considers practical aspects of actigraphy in expert ABPM for clock-independent calculation...

Combined gemcitabine, cisplatin, dexamethasone, methotrexate, and pegaspargase (GDP-ML) for patients with newly diagnosed extranodal natural killer/T cell lymphoma, nasal type: a single arm, single center, prospective phase 2 study
Abstract Extranodal natural killer/T cell lymphoma, nasal type (ENKL) is a highly aggressive tumor with relatively poor prognosis. In this prospective study, we investigated the efficacy and toxicity of a novel GDP-ML regimen (combined gemcitabine, cisplatin, dexamethasone, methotrexate, and pegaspargase) as front-line treatment in newly diagnosed ENKL. Eligible newly diagnosed stage I/II ENKL patients received sandwich chemoradiation therapy. Patients with stage III/IV disease...

Clinical Utility of Reflex Ordered Testing for Molecular Biomarkers in Lung Adenocarcinoma
Metastatic lung adenocarcinoma patients may not be getting the correct molecular biomarker testing prior to first line therapy. We implement reflex ordered testing for molecular biomarkers at time of pathologic diagnosis of lung adenocarcinoma. We retrospectively compared reflex ordered testing to standard molecular testing. Reflex ordered testing lead significantly shorter turn around time and higher variant detection rate.

Graphitic carbon nitride–doped sewage sludge as a novel material for photodegradation of Eriochrome Black T
Abstract The bio-resource utilization of sewage sludge is presented by preparation of novel waste sludge–doped graphite carbon nitride (g-C3N4) photocatalyst. The sludge flocs which constitute bacteria and organic substances served as a pore-forming framework in the catalyst, while the inorganic fractions including those transition metals and crustal metals can function as dopants for sludge-based g-C3N4 composite. The physicochemical properties of as-prepared catalyst were well...

Hypersensitivity reactions to biologics (part I): allergy as an important differential diagnosis in complex immune-derived adverse events
Summary Purpose Biotechnological substances (BSs) are strongly relied upon to prevent rejection of transplanted organs, and to treat oncological, allergological, and other inflammatory diseases. Allergic reactions to partly foreign biologics can occur due to their potential immunogenicity. The severity of an immune response to a biological drug may range from no clinical significance to a severe, life-threatening anaphylactic reaction....

Adverse outcome pathways for ionizing radiation and breast cancer involve direct and indirect DNA damage, oxidative stress, inflammation, genomic instability, and interaction with hormonal regulation of the breast
Abstract Knowledge about established breast carcinogens can support improved and modernized toxicological testing methods by identifying key mechanistic events. Ionizing radiation (IR) increases the risk of breast cancer, especially for women and for exposure at younger ages, and evidence overall supports a linear dose–response relationship. We used the Adverse Outcome Pathway (AOP) framework to outline and evaluate the evidence linking ionizing radiation with breast cancer from...

Bifurcated binding of the OmpF receptor underpins import of the bacteriocin colicin N into Escherichia coli [Membrane Biology]
Colicins are Escherichia coli–specific bacteriocins that translocate across the outer bacterial membrane by a poorly understood mechanism. Group A colicins typically parasitize the proton-motive force–linked Tol system in the inner membrane via porins after first binding an outer-membrane protein receptor. Recent studies have suggested that the pore-forming group A colicin N (ColN) instead uses lipopolysaccharide (LPS) as a receptor. Contrary to this prevailing view, using diffusion-precipitation...

A tri-modular bacterial enzyme combining hydrolytic activity with oxidative glycosidic bond cleavage efficiently degrades chitin [Enzymology]
Findings from recent studies have indicated that enzymes containing more than one catalytic domain may be particularly powerful in the degradation of recalcitrant polysaccharides such as chitin and cellulose. Some known multi-catalytic enzymes contain several glycoside hydrolase (GH) domains and one or more carbohydrate-binding modules (CBMs). Here, using bioinformatics and biochemical analyses, we identified an enzyme, Jd1381 from the actinobacterium Jonesia denitrificans, that uniquely combines...

Structural optimization of a TNFR1-selective antagonistic TNF-{alpha} mutant to create new-modality TNF-regulating biologics [Protein Structure and Folding]
Excessive activation of the proinflammatory cytokine tumor necrosis factor-α (TNF-α) is a major cause of autoimmune diseases, including rheumatoid arthritis. TNF-α induces immune responses via TNF receptor 1 (TNFR1) and TNFR2. Signaling via TNFR1 induces proinflammatory responses, whereas TNFR2 signaling is suggested to suppress the pathophysiology of inflammatory diseases. Therefore, selective inhibition of TNFR1 signaling and preservation of TNFR2 signaling activities may be beneficial for managing...

Tripartite-motif family protein 35-28 regulates microglia development by preventing necrotic death of microglial precursors in zebrafish [Cell Biology]
Microglia are tissue-resident macrophages in the central nervous system (CNS) that play essential roles in the regulation of CNS development and homeostasis. Yet, the genetic networks governing microglia development remain incompletely defined. Here, we report the identification and characterization of a microglia-defective zebrafish mutant wulonghkz12 (wulhkz12) isolated from an ethylnitrosourea (ENU)-based genetic screen. We show that wulhkz12 mutants harbors a missense point mutation in the gene...

Novel role of ASH1L histone methyltransferase in anaplastic thyroid carcinoma [Molecular Bases of Disease]
Anaplastic thyroid cancer (ATC) is one of the most aggressive human malignancies, with an average life expectancy of ~6 months from the time of diagnosis. The genetic and epigenetic changes that underlie this malignancy are incompletely understood. We found that ASH1-like histone lysine methyltransferase (ASH1L) is over-expressed in ATC relative to the much less aggressive and more common differentiated thyroid cancer. This increased expression was due at least in part to reduced levels of microRNA-200b-3p...
