- Generation of Porcine Testicular Organoids with Testis Specific Architecture using Microwell CultureHere, we present a protocol for the reproducible generation of porcine testicular organoids with testis specific tissue architecture using the commercially available microwell culture system.
- Bacterial Peptide Display for the Selection of Novel Biotinylating EnzymesHere we present a method to select for novel variants of the E. coli biotin-protein ligase BirA that biotinylates a specific target peptide. The protocol describes the construction of a plasmid for the bacterial display of the target peptide, generation of a BirA library, selection and characterization of BirA variants.
- Dissection, Culture and Analysis of Primary Cranial Neural Crest Cells from Mouse for the Study of Neural Crest Cell Delamination and MigrationThis protocol describes the dissection and culture of cranial neural crest cells from mouse models, primarily for the study of cell migration. We describe the live imaging techniques used and the analysis of speed and cell shape changes.
- Applications for Open Source Microplate-Compatible Illumination PanelsMicroplate Assistive Pipetting Light Emitter (M.A.P.L.E.) is a computer-driven device that systematically illuminates microtiter wells to provide guidance for the manual preparation of microplates. M.A.P.L.E. improves the accuracy of microplate preparation while automating data recordkeeping. In addition, it can assist with examining microplate quality or aid in the detection of errors.
- Lentiviral CRISPR/Cas9-Mediated Genome Editing for the Study of Hematopoietic Cells in Disease ModelsDescribed are protocols for the highly efficient genome editing of murine hematopoietic stem and progenitor cells (HSPC) by the CRISPR/Cas9 system to rapidly develop mouse model systems with hematopoietic system-specific gene modifications.
- Precision Implementation of Minimal Erythema Dose (MED) Testing to Assess Individual Variation in Human Inflammatory ResponseMinimal erythema dose (MED) testing is used to establish dosage schedules for ultraviolet radiation phototherapy. It can assess individual variation in inflammatory response but lacks methodology for achieving reproducible results. Here, we present a precision implementation of MED and demonstrate its ability to capture individual variation in inflammatory response.

- Printing Custom Flexible Electronics Directly Onto Skin, Bandages, Medical DevicesFlexible electronics have the potential to help monitor, and even modulate, a number of physiological parameters. While stick-on heart monitors can be manufactured as one-size-fits-all, in applications such as electronic bandages that can monitor and treat a wound, it’s best to be able to create custom devices that suit each patient’s needs. Currently, making flexible electronics requires harsh processing, including chemical baths, hardeni
- Laser Activated Gold Nanorods Create Silk Seal for Incisions and WoundsStaples and sutures are currently used to seal soft tissues together in clinical practice, but researchers at Arizona State University have come up with an alternative technique that resembles welding. It may end up being used to seal tissues during surgeries and to treat wounds, and in many cases simply to enhance the effectiveness of staples and sutures. Moreover, because the seal can be more continuous, covering the entirety of an incision, it
- New Imaging Technique Reveals Deep Tumors, Helps Attack ThemTumors deep inside the body can be very difficult to spot, track, and study. The brain, being surrounded by a thick skull, is particularly challenging to image using light, so MRI and CT are currently the go-to imaging modalities when looking at deeply seated tissues. Now, researchers at Stanford University are reporting the development of new nanoparticles that can be used to light up and image tumors located well below the surface of th

- Treatment Protocol for Compromised Nasal SkinAs the number of patients seeking surgical and nonsurgical rhinoplasty continues to increase, the risk of nasal skin compromise after surgery also has risen. Vascular insult to the nasal skin envelope can lead to permanent disfigurement that is nearly impossible to correct. Tissue loss often requires major reconstruction that yields suboptimal cosmetic results. This article discusses prevention, early recognition, and effective treatment that aim
- Filler-Associated Vision LossSoft tissue fillers continue to gain popularity in addressing volume loss and changes associated with facial aging. The rare but devastating complication from iatrogenic vascular occlusion can result in irreversible vision loss. This article discusses the complications of vision loss associated with fillers and reviews applicable treatment techniques and prevention methods.
- Management of Postsurgical Empty Nose SyndromeEmpty nose syndrome (ENS) is a controversial condition associated with disruption of nasal airflow caused by excessive loss of turbinate tissue. ENS arises after total or near-total inferior turbinate resection. Patients present with intense fixation on the perception of nasal obstruction. Diagnostic tools to assess for empty nose syndrome include a validated patient questionnaire and the office cotton test. Treatment involves topical moisturizat
- Neck Deformities in Plastic SurgeryThis article provides facial plastic surgeons with the insight to avoid and address common pitfalls in neck procedures. Many aesthetic issues are created from overtreatment or undertreatment of components of the neck. Using the platysma muscle as the divide, ease of access to superficial anatomy leads to overtreatment problems, whereas difficulty of access to deeper structures leads to undertreatment problems and to overall imbalances. - Revision Facial Plastic Surgery: Correcting Bad ResultsAll facial plastic surgery procedures and treatment modalities have potentially unintended outcomes, possible poor results, or complications. These issues should be minimized through proper preoperative planning, patient selection, and careful technique. However, even in the experience of well-trained and appropriately careful surgeons, complications can and do occur. Planning for correction of these unexpected and unfortunate situations should b
- Management of Surgical ScarsAn ideal scar is flat, thin, and color matched to the surrounding skin. Incision planning, skin closure, and postoperative care are vital to create an inconspicuous scar. Depressed, hypertrophic, and keloid scars each pose unique challenges to the facial plastic surgeon. Several surgical and nonsurgical options exist in the treatment of scars. Appropriate treatment is based on scar location, quality, and size as well as patient history, preferenc
- Tips to Avoid Complications Following Mohs ReconstructionTo avoid complications or bad results following reconstruction of skin cancer defects, there are several factors that can be incorporated into the design and execution of repairs. These techniques not only decrease the need to correct bad results, they improve the final aesthetic and functional outcome.
