Clinical utility of plasma cell-free DNA in adult patients with newly diagnosed glioblastoma - a pilot prospective study
Purpose: The clinical utility of plasma cell-free DNA (cfDNA) has not been assessed prospectively in patients with glioblastoma (GBM). We aimed to determine the prognostic impact of plasma cfDNA in GBM, as well as its role as a surrogate of tumor burden and substrate for next-generation sequencing (NGS). Experimental Design: We conducted a prospective cohort study of 42 patients with newly diagnosed GBM. Plasma cfDNA was quantified at baseline prior to initial tumor resection and longitudinally during...

Understanding the Right to Try Act
Cancer patients who have exhausted standard treatments often seek access to investigational drugs. Often, however, such access is unavailable, due to either the unavailability of a trial, lack of an open recruiting spot on the trial, even when the trial itself is open, or the inability of the patient to meet one or more trial eligibility criteria. In such settings patients often seek access to investigational agents outside of a trial. The federal "Right to Try" legislation was passed to create...

Targeting of {beta}-catenin reverses radioresistance of cervical cancer with the PIK3CA E545K mutation
This study aims to explore whether E545K, the most common hot-spot mutation of PIK3CA in cervical cancer, confers radioresistance to cervical cancer cells, to demonstrate the underling mechanism and to develop the effective targets. SiHa and MS751 cells with PIK3CA-WT and PIK3CA-E545K were established by lentiviral transfection. The radiosensitivity was assessed by colony formation, cell cycle, cell apoptosis, DNA damage and repair assay. The growth and immunohistochemistry assay of xenograft tumor,...

Statins use and prostate cancer mortality
The association between statins therapy and prostate cancer is extensively studied [1,2]. Recently, one cohort study conducted by Van Rompay et al. published in European Journal of Cancer demonstrated that statins use was associated with a lower mortality of prostate cancer than non-use (hazard ratio 0.73, 95% confidence interval 0.66–0.81) [3]. This result was compatible with another cohort study in Finland showing that statins use after prostate cancer diagnosis was associated with 20% reduced...

Polymorphisms in oxidative stress pathway genes and prostate cancer risk
Abstract Purpose Age-related factors including oxidative stress play an important role in prostate carcinogenesis. Can synthetic lethality approach be used with DNA repair genes for primary and secondary MDS?
Abstract Cancer-specific defects in DNA repair pathways create the opportunity to employ synthetic lethality approach. Recently, GEMA (gene expression and mutation analysis) approach detected insufficient expression of BRCA or NHEJ (non-homologous end joining) to predict PARP inhibitors response. We evaluated a possible role of DNA repair pathways using gene expression of single-strand break (XPA, XPC, XPG/ERCC5, CSA/ERCC8, and CSB/ERCC6) and double-strand...

Abstract Purpose Recently, the potential value of isocitrate dehydrogenase (IDH) mutation as a prognostic marker in glioblastomas has been established. Glioblastomas are classified by their IDH mutation status under the 2016 WHO classification system. However, noninvasive diagnostic methods for the mutation status in glioblastoma patients have not been established so far. The purpose of this study was to evaluate the difference of acetate...

Ki67 expression and localization of T‐cells after neoadjuvant therapies as reliable predictive markers in rectal cancer
Abstract Chemoradiotherapy (CRT) is the standard neoadjuvant therapy for locally advanced rectal cancer (RC). However, neoadjuvant chemotherapy (NAC) also shows favorable outcomes. Although the immunological environment of RC has been thoroughly discussed, the effect of NAC on it is less clear. Here, we investigated the immunological microenvironment, including T‐cell infiltration, activation, and topological distribution, of resected RC tissue after neoadjuvant therapies and evaluated the correlation...
